每天都会上小木虫瞅瞅，貌似已经成为一种习惯了，但是最近天然药化版块好像被“微谱求助”占领了（我并不反感“微谱求助”，自己也求助过，但是总觉得求助结果好像仅仅对求助者有参考价值，所以在想版主大人能不能为它辟出一个独立的位置！扯远了！），所以突发奇想，想跟各位同仁交流一下各位收藏的天然药化方面的经典文献！当然，我称之为“经典”的，并非是那些以化合物的新颖度或强大活性为亮点的文献（尽管现在很多搞天然药化的都是这个思路），而是那些能给我们的研究提供启示，或是能够提供新颖的研究思路的好文章！希望各位虫友们（特别是那些广读文献的虫友）能不吝拿出自己“压箱底”的文献，让我们在交流中进步（需要下载权限的文献麻烦大家上传个附件吧）。

文献亮点：本文提出了一个核心的思路—“Chemically engineered extracts”（我理解为“提取物的化学改造”）。研究者发现，在药物开发方面，天然产物相对于合成产物具有很多优势，而与此同时，现有药物及利用组合化学获取的分子也具有一些自身特点，如它们的结构中有更多含氮原子、硫原子、卤原子的官能团。于是研究者设想，如果能将这些官能团引入到天然产物中，会不会是对大自然制造天然产物的化学能力的一种补充和强化呢。于是研究者萌生了对某植物提取物中化合物进行“集体改造”的想法，从DNP中统计得知，天然产物中最常见的官能团为羰基，因而被作为引入特殊官能团的切入点，同时，他们选择了可与各种类型羰基发生反应的肼作为对提取物进行“改造”的“武器”，结果使原本不存在抗菌活性的提取物产生了抗菌活性。通过活性追踪的方法，研究者从“改造”后的提取物中分离得到了具有抗菌活性的化合物，并证实了该化合物为原提取物中化合物与肼反应所得产物。
评注：这种思路有点像童话里的王子找灰姑娘的办法—给每个姑娘都试穿水晶鞋，最后总有一个穿着合脚，那个就是灰姑娘。很多天然产物分子或许也是灰姑娘，但是需要穿上“水晶鞋”才能成为“公主”。此外，该文章研究者后面还开展了一些相关研究，并于2011年在“Accounts of Chemical Research”上发表了题为“Chemically Engineered Extracts: Source of Bioactive Compounds”的文章（类似综述）。我个人觉得，对于我们自身而言，或许可以依据自己研究的化合物的结构特点进行一些针对性的“改造”，以发现有价值的分子。
