【求助/交流】跪求用alamar blue检测hl60细胞毒性的方法
各位大虾，小女子跪求用alamar blue检测hl60细胞毒性的方法。
我是细胞实验方面的新手，现导师要求我用hl60筛选细胞毒性，用alamar blue的方法。我做了2次了，都是颜色变化不大。
具体实验是这样做的： 细胞播种10×4个在96孔板，加入我的提取物24，48，72小时后
用酶标仪测吸光值。测吸光值之前6小时，加入alamar blue。即便是72小时候，颜色变化都不大，基本还是蓝色。这样无法计算细胞毒性。我的提取物是没有问题的，在做其他的细胞筛选是都显示出了良好的细胞毒性。
我不知道为什么颜色没有变化，跪求有相同实验经验的大虾指教。
如能帮我解决问题，即送30金币。
个人猜测原因可能有两个方面：
一. alamar blue与细胞孵育的时间不够。参见alamar blue使用说明：
1. 细胞的增殖和细胞毒实验，一般可在96孔细胞培养板中进行。& &
& & 2. 每孔加入100微升细胞悬液。细胞的数量取决于实验目的和培养时间。增殖实验每孔通常加入103个以上数量的细胞；细胞毒性实验每孔至少加入5x103个或以上数量的细胞(具体每孔所用的细胞的数目，需根据细胞的大小，细胞增殖速度的快慢等因素确定)。& &
& & 3. 接种的细胞按照实验需要，送入细胞培养箱进行培养。增殖实验通常要给予0-10微升特定的药物或生长因子进行刺激。细胞毒实验通常要给予0-10微升抑制因子或细胞毒药物；& &
& & 4．培养结束后，取出阿尔玛蓝试剂，置室温融化混匀，于洁净工作台内按10微升/孔加入微孔板中，在细胞培养箱内继续孵育12－24小时，培养液颜色由蓝变为粉红（如采用荧光分光光度法，只需孵育5－8小时）；& &
& & 5．在570nm测定吸光度，参考波长600nm。如无此滤光片， 可用565 nm 和 610 nm的滤光片替代。&&
& &时间太短，alamar blue作为氧分子电子传递链受体的功能未充分发挥。楼主可尝试孵育24小时。
二. 如果楼主使用的hl60细胞增殖太快，那也可能导致不同时间处理组差异不明显。因为细胞增殖过快，平均到每个细胞的细胞毒效果也就弱化了。所以才会要求接种之时，细胞密度要适宜。
另外我觉得，如果用alamar blue效果不好的话，可以尝试下MTT法嘛。我也做过化合物的细胞毒性试验，用MTT效果一直都不错的。 Originally posted by henkally at
